L-theanine crosses the blood-brain barrier and increases levels of dopamine and serotonin the brain. The mode of action includes increasing GABA activity in the brain as well.
Caffeine works by essentially blocking adenosine (a receptor responsible for making you sleepy) from binding.

The toxicity studies on caffeine demonstrate that it is relatively safe for human consumption; this is of course not without it limits because there are instances of caffeine overdose. A lethal dose of caffeine is said to be around 10 grams.
